James Taylor Wise (a civil surgeon in Dhaka) founded it in 1835 as an English seminary school (now Dhaka Collegiate School). Mr. Wise, with the assistance of the District Magistrate, Mr. Grant, formed a local Committee of Public Instruction. The school building was built partly from public donations on the site of an English factory.

The College granted permission for the school on July 18, 1841. The foundation stone of the institution was laid on November 20, 1841, and the construction was completed in 1846 with the help of the Bishop of Calcutta. The first graduating class included Muslim and Hindu students, as well as a number of foreign students, mainly from Armenia and Portugal.

To accommodate the physics and chemistry facilities, the institution was moved to a large site east of Victoria Park in 1873. It moved to Curzon Hall in 1908, while the science departments were housed in the present chemistry building of the University of Dhaka, which also included two new halls.

The college moved to the old High Court building in 1921 when the University of Dhaka was established there. During the Second World War, it was shifted to Purana Dhaka’s Siddique Bazar. In 1955, the college established its own permanent location at Mirpur Road, Dhanmondi, near Dhaka New Market.
