James Taylor Wise (a civil surgeon in Dhaka) founded it in 1835 as an English seminary school (the current Dhaka Collegiate School). Mr. Wise, with the assistance of district magistrate Mr. Grant, formed a local Committee of Public Instruction. The school building was built partly out of public donations on the grounds of an English factory.

The college granted the school approval on July 18, 1841. The foundation stone for the institution was laid on November 20, 1841, and construction was finished in 1846 with the assistance of the Bishop of Calcutta. There were Muslim and Hindu students in the first graduating class, as well as a number of foreign students, primarily from Armenia and Portugal.

In order to accommodate the physics and chemistry facilities, the institution was transferred in 1873 to a spacious facility east of Victoria Park. It relocated to Curzon Hall in 1908, while the science departments were housed in the University of Dhaka’s current chemistry building, which also included two new halls.

The college relocated to the old High Court Building in 1921 when the University of Dhaka was established there. During WWII, it was relocated to Purana Dhaka’s Siddique Bazar. In 1955, the college established its own permanent site on Mirpur Road, Dhanmondi, near Dhaka New Market.
